Manufacturing of Phage Lysin

Creative Biolabs has years of experience in the field of phage with advanced technology and extensive experience. In light of the threat of antibiotic resistance, the bacterial cell wall hydrolase, phage lysin, encoded by a double-stranded DNA phage, has been progressively developed as a novel antibacterial product for the microbial industry, clinical practices, and biotechnology. We provide bioengineering services for phage lysin as well as small-scale and large-scale manufacturing services. With our world-leading manufacturing technologies, our custom development and manufacturing services will accelerate the progress of groundbreaking research related to phage lysin.

Production Host Systems Identity Confirmation

First, we customize your phage lysin according to your needs according to different modification or engineering strategies, including mutagenesis and protein truncation, domain shuffling and fusion, PEGylation, glycosylation, and T-cell epitope depletion, etc. Different cloning hosts that can be used for recombinant protein production include bacteria, yeasts, insect cells, mammalian cells, filamentous fungi, transgenic animals, and transgenic plants. We provide Escherichia coli strains as hosts for recombinant protein overproduction and culture them in lysogeny broth (LB) medium. The quality and purity of the strains used for production are guaranteed under our quality control. Before recombinant protein expression, we fuse the modified phage lysin gene into the plasmid and transformed it into the host strain and confirm it by double restriction digestion analysis and automated DNA sequencing.

Recombinant Protein Expression

We use E. coli containing recombinant plasmids as hosts for recombinant protein production, and culture bacterial cells in LB medium at 37°C or lower temperature to avoid precipitation with shaking and appropriate adding of lactose and other nutrients that facilitate E. coli recombinant protein production. Our excellent temperature control and large-scale culture facilities allow the transcribed recombinant proteins to fold correctly into their active form while ensuring high titer and purity of phage lysins.

Phage Lysin Purification

Purification procedures for phage lysis proteins are essential to remove contaminants and impurities while ensuring the correct tertiary structure and lytic activity of the proteins. Creative Biolabs designs and provides purification services tailored to different phage lysins. We use the following phage lysin purification processes or combination of multiple purification processes depending on different phage lysins:

  • Ion-exchange chromatography
  • Affinity chromatography
  • Immobilized metal ion affinity chromatography (IMAC)
  • Size exclusion chromatography
  • Expanded bed adsorption
  • Gel filtration
  • Ultrafiltration

Stabilization and Formulations

In the initial design of the engineered phage lysins, we have considered the phage lysin stability. Site-direct mutagenesis by computational modeling can be used to enhance the thermal stability of phage lysins. To achieve optimal therapeutic efficacy of the final drug product of phage lysins with guaranteed safety and stability, our formulation services, including liquid formulation, lyophilization, encapsulation, emulsification, or microencapsulation services, ensure the balance between adjuvant and lytic protein, protecting protein particles from aggregation, adsorption, and denaturation.
