Background

The cell walls of Gram-positive bacteria are rich in carbohydrate structures. In general, carbohydrates elicit a poor immune response. The lysibody approach provides new directions to address this shortcoming. As an engineered molecule, lysibodies combine a phage-derived high-affinity domain and the Fc effector portion of a human IgG antibody. Evidence suggests that lysibodies are closely linked to the treatment and prevention of infection with drug-resistant pathogens.

Significance of Binding Range Determination Assay

In general, it is necessary to characterize all lysibodies to confirm their therapeutic potential. The most important step is to examine the binding capacity to specific bacterial strains and the host range of the lysibodies. Characterizing the host range could also test whether the lysibody could be used to treat infections with other pathogenic strains, making it easier to find the most effective method.

Principle and Procedures

At Creative Biolabs, assays relying on fluorescence microscopy to test lysibody binding to the cell walls of different bacteria are available. Fluorescent dyes are frequently used in detection. The novel dyes we employed are very lightfast and have high fluorescence quantum yields. Furthermore, red-emitting fluorescent markers are very valuable in biological microscopy because they minimize cellular autofluorescence and increase the flexibility of multicolor experiments. In the assay, firstly, the strain to be detected needs to be selected. Bacterial cells are then fixed, attached to microscope coverslips, and blocked. Cells are then incubated with a specific lysibody followed by an anti-human IgG Fc fluorescence conjugate. According to the results, the lysosomal host binding range is determined.
