Lysibody-induced Complement Deposition Determination Assay

In recent years, the role of lysibody in inducing complement deposition has attracted widespread attention. As an expert in the field of lysibody, Creative Biolabs is committed to providing global customers with a full range of lysibody-induced complement deposition determination assay services, from experimental design to result analysis.

Overview of Lysibody

The cell walls of Gram-positive bacteria contain abundant carbohydrate structures, however, the poor immune response to carbohydrates limits their function. The advent of the lysibody provides an attractive tool to address this challenge. As an engineered molecule, lysibodies combine a phage-derived binding domain with the Fc portion of a human immunoglobulin G (IgG) antibody. Thus, lysibodies are closely linked to the efficient immunity of bacterial surface carbohydrates. Furthermore, the Fc moiety attracts complement and professional phagocytes and promotes bacterial clearance in vitro and mouse sepsis models.

Principles and Procedures

Deposition of complement on the surface of pathogens is an important mechanism for their clearance by phagocytes. In general, fluorescence microscopy is a crucial tool to determine whether lysibodies can induce complement fragment fixation on the surface of Gram-positive bacteria. Lysibodies play a crucial role in inducing complement deposition on the cell surface of Gram-positive bacteria. The cells of Gram-positive bacteria are attached to specially treated coverslips and incubated with lysibodies. The cells are then incubated with human complement, washed, fixed, and blocked. The experimental results are presented by microscopy using a C3-specific antibody and a fluorescent conjugate to assess the extent of complement deposition on the Gram-positive bacteria.
